The highly sought-after details will allow fans to line the streets and farewell the team before Sunday's grand final in Sydney.
The bus will depart the Knights' centre of excellence in Broadmeadow at 9.30am.
It will head west along Lambton Road, turn right onto Turton Road and head past the club's home ground, McDonald Jones Stadium.
From there, all roads lead to the freeway along Griffiths and Newcastle road past Lambton, Waratah and Jesmond.
The bus will veer left onto Thomas Street at Wallsend, where large crowds are expected to gather alongside the touch football fields.
It will then proceed along Newcastle Link Road to the M1 Motorway, previously known as the F3 Freeway, where it will turn southward for the journey to Sydney.
Thousands of fans are expected to see off the players, much like before the club's 1997 and 2001 grand finals.
The club has requested fans gather roadside along the route, rather than at the centre of excellence.
